Abstract

Infection is a process of invasion and breeding of microorganisms that occur in human tissue which can cause local cellular injury, one of the infectious diseases causes by bacteria E.coli. Detection of bacteria thet cause infectionon human body with conventional methods usually takes 24 hours after examination the symptoms of infection. This study has a purpose to know the ability of gas chromatography to analyze the compound product from E.coli bacteria at growth time less than 24 hours. Second, to know different of chromatogram from compound product of E.colibacteria a various incubation time, and third, to find how long incubation time of E.coli bacteria that can identify the compound with GC. Derivatization the fatty acid of E.coli is transform to fatty acid metal ester (FAME) by esterification method. Variations in the growth time of E.coli bacteria are perform at 6, 12, 24, 36 and 48 hours, respectively. E. coli bacteria samples that will separate the fatty acid and esterified into fatty acid methyl ester. Identification of E.coli bacteria was analyze by gas chromatography with the initial temperature conditions of column 1200C. In the analysis using gas chromatography all variations of bacterial growth time can be identified. The retention time (tR) obtained from each sample was 3.737; 3,795; 3,477; 3,830; and 3.470 minutes. The results of this study revealed that gas chroatography was able to identify bacteria at growth time of less than 24 hours, was 6 hours and 12 hours, and the most optimal time for gas chromatography can identify E.coli bacteria was 6 hours growth.
