Abstract
Loratadine is a tricyclic antihistamine and an azatadine derivative, the effect of loratadine do not show a clinically significant sedative effect on dosing of 10 mg. However, Loratadin has low bioavailability, and insoluble in water. So, the purpose of this research is to formulate the Loratadin in the Self- Nanoemulsifying Drug Delivery System (SNEDDS) dosage form, which is a nanoemulsion dosage form comprising a mixture of drugs, oils carrier, surfactants and cosurfactants. This research method uses experimental design which is done by formulating SNEDDS dosage form using different surfactant and co-surfactant ratio. From all of the homogeneous dosage form then the researcher made the observation of transmittance value and time emulsification to choose the best supply. After that, the researcher did some tests on the best formula to determine the characterization. The tests includes the observation of particle size and the distribution of particle size, and drug loading. And after that the researcher performed stability test by centrifugation method and Freeze Thaw Cycle. The best dosage form results of SNEDDS Loratadin has a Cremophor RH40 (surfactant) ratio: propylene glycol (co-surfactant) 1: 1 and 1: 2. Transmit rates above 10% with time emulsification less than 1 minute. The SNEDDS supply may form the nano emulsions in water with particle sizes within the range of 20-200 nm and have sufficient particle distribution to satisfy the criteria as nanoemulsion.
