Abstract

Interactive mixture occurs because of the interaction (attachment) between the drug component (micronized) with the granule of the host (host) is much larger the size of the particles. Small doses of drugs such as prednisone prepared by interactive mixed techniques can ensure homogeneity and stability, and can speed up the dissolution of the drug. In addition, because the carrier particles are relatively coarse, the properties of flow and compressibility are relatively good, so they can be made in direct printing. Physical properties of tablets including hardness, brittleness and disintegration and dissolution times are influenced by, among others, fillers. Therefore, a combination of fillers was done on the preparation of mixed prednisone interactive tablets. The experiment was conducted with three formulas: formula I, II and III, respectively using avicel PH 101: lactose (100: 0), avicel PH 101: lactose (50:50) and avicel PH 101: lactose (0: 100). The three formulas tested for physical properties include weight uniformity, hardness, brittleness, disintegration time, uniformity of content and dissolution. The results data are then compared to the applicable requirements. The optimum formula is determined by simplex lattice design. The difference of the three formulas was analyzed by one way anova method with 95% confidence level. The result of this research showed that the optimum formula is avicel PH 101: lactose 71,7mg: 28,3 mg with fragile response 0,78%, crushed time 5,52 minute and dissolution 132,81% which mean that between optimum formula with prediction at Simplex lattice design shows no significant difference in fragile response, disintegration time and dissolution.
