Abstract
Loratadine is a second generation long-acting antihistamine drug that works long with antagonistic activity against selective H1 histamine peripheral receptors. The solubility of loratadine is low in water so that nanosphere preparations can be made to increase the bioavailability of the drug in the body. The aim of this research is to know the combination of polyvinyl alcohol polymer with tween 80 with sonication emulsification method capable of producing nanosphere preparation. This study used three formulas consisting of a combination of loratadine, polyvinyl alcohol and tween 80 with the sonication emulsification method and characterization of particle size, entrapment efficiency, and zeta potential. The results showed the best characterization of nanosphere loratadine based on particle size ie nanosphere loratadine formula 2 with a time of 15 minute sonication with particle size of 145.20 ± 0.01 nm, the efficiency of 11.31% and zeta potential of -3.98 mV, while the best formula based on the entrapment efficiency of nanosphere loratadine formula 3 with 15 minute sonication with particle size of 173.80 ± 0.01 nm, 13.00% and zeta potential of -3.88 mV.
