Abstract

Piroxicam is a nonsteroid anti-inflamation (AINS) used as analgesics and antiinflammation in rheumatoid arthritis, osteoarthritis, and acute pain in musculoskeletal disorder. This drug). This research aimed to find out the effect of omeprazole administration on pharmacokinetic profile of piroxicam in New Zealand male rabbit and to find has irritating effect on gastrointestinal tract. The treatment of gastritis due to non-steroid anti-inflammation drug (OAINS) is done so far using omeprazole to inhibit the proton pump transporting H+ out of gastric pariental cell (proton pump inhibitorout the effect of per oral omeprazole administration on pharmacokinetic absorption, distribution, and elimination phases of piroxicam in New Zealand male rabbit. The tested animal was divided into 3 groups, each of which consisting of 3 tested animals. The first group was positive control given piroxicam suspension, negative control given CMCNa 1% and treatment group given piroxicam and omeprazole suspension. The blood was taken using marginalis vein everyday for 11 days with sampling time at hours 12, 24, 36, 48, 60, 72, 84, 96, 108, and 120. Piroxicam level of blood was determined using UV-Vis spectrophotometer. The result of research and statistic test showed that omeprazole affected the pharmacokinetic profile of piroxicam. From the result of research, it could be found that per oral omeprazole improved the pharmacokinetic score of Ka, Cpmax, t½, but did not affect pharmacokinetic profile of piroxicam significantly in New Zealand male rabbit.
