Abstract
Ketoprofen is a non-steroidal anti-inflammatory drug (NSAID) used in musculoskeletal and joint disorders such as ostheoarthritis and rheumatoid arthritis. Ketoprofen belongs to the second class BCS (Biopharmaceutics Classification System) drug group, with high membrane permeability and low solubility. The inclusion complex of ketoprofen in β-cyclodextrin is used to increase solubility and mask the taste. Formulations in the form of FDT (Fast Disintegrating Tablet) preparations can improve patient comfort. This research aimed to determine the effect of superdisintegrant combination of Crosscarmellose Sodium and Sodium starch glycolate on physical quality and dissolution profile. The inclusion complex was prepared by kneading method with 1:1 molar ratio and characterized using Fourier - Transform Infrared (FTIR) spectroscopy and Differential Scanning Calorimetry (DSC). Tablets were prepared by directpress method with variation concentration of Crosscarmellose sodium and Sodium starch glycolate with ratio 100:0%, 50:50%, 25:75%, 75:25%, 0:100%. Tests conducted to determine the physical quality of ketoprofen FDT are hardness, fragility, wetting time, crushed time, and dissolution. The results showed that the combination of superdisintegrant Croscarmellose sodium and Sodium starch glycolate (25: 75%) had an effect on the physical quality and release of the best ketoprofen FDT drugs.
