Abstract

Prednisone have bad solubility in water that affect the bioavailability in body. The interactive mixture method is one of the ways to accelerate the solubility of the drug by placing the drug in micronized form attached to the host (the carrier). The purposes of this research is to know the influence of the combination of filler Avicel PH 101 and lactose as a host in the creation of interactive mixture prednisone tablets. The optimum proportions of filler material Avicel PH 101 and lactose will give influence on the physical quality and profile dissolution tablets of the prednisone interactive mixture tablets. This research was conducted with 3 variations with the proportion of filler material 100% (93,5 mg) Avicel PH 101 : 0% (0 mg) lactose; 50% (46,75 mg) Avicel PH 101 : 50% (46,75 mg) lactose and 0% (0 mg) Avicel PH 101 : 100% (93,5 mg) lactose. Each formula is done the process of tablet and carried out testing against a critical point of fragility, time of disintegration and dissolution. The results are then carried out an analysis of the Simplex Lattice Design to gain optimum formula. The results of this research obtained optimum formula i.e. proportion of Avicel PH 101 greater than lactose that is of 80,784 mg : 12,716 mg, test the fragility of 0,45%, time of disintegration is 4,40 minutes and better dissolution Q30 takes 143,05%.
