Abstract
Fisetin belongs to BCS class II with low solubility and high permeability. Fisetin has very low bioavailability of 10%, due to its small water-solubility (0.002 mg/ml) and low absorption so that the administration of fisetin in oral and dermal preparation is limited. Fisetin solubility can be improved using drug delivering system such as transfersome. This research aimed to find out whether or not fisetin can be made transfersome using thin film hydration method, the effect of surfactant concentration on the entrapment efficiency of transfersome, the characterization of transfersome and the stability of fisetin transfersome. Transfersome was prepared using thin film hydration method because simple preparation results in elastic and flexible nano vesicle. In this research, varying concentration ratios of phosphatidilcholine: span 80 were given: 475mg:25mg in formula 1; 462.5mg:37.5mg in formula 2; 450mg:50mg in formula 3; 425mg:75mg in formula 4; 400mg:100mg in formula 5. Each formula used fisetin 10 mg as active substance. Fisetin was made transfersome in nanometer size using thin film hydration method. Vesicle produced was then characterized. The characterization of fisetin transfersome included particle size, particle morphology, and entrapment efficiency. Fisetin could be made transfersome using thin film hydration method; the different concentration of surfactant resulted in different efficiency of transfersome absorption. The characterization of fisetin transfersome resulted in particle sizes of 332.43; 593.53; 620.9; 671.43; 412.8nm in F1, F2, F3, F4 and F5, respectively. The entrapment efficiency in the five formulas was 73.01%, 82.75%, 86.64%, 90.34%, and 92.43%, respectively. From SEM and stability tests conducted on formula 5, it could found transfersome with irregular shape and particle size of 562.46 nm and potential zeta of 0.19 mV. Fisetin transfersome in formula 5 experienced a slight increase in its particle size but it was stable enough.
