Abstract
Angkak (red yeast rice) is a product of fermented rice by the mold Monascus purpureus which contains anthocyanin compound of flavonoids class. Angkak extract has high antioxidant activity. Improvement of delivery system by creating nanoemulsion preparation is needed to increase antioxidant activity. The easy method to apply nanoemulsion of angkak extract is by turning it into cream form. This research aims to find angkak extract’s antioxidant activity and its formulation in form of both nanoemulsion and cream, which will be further tested to find its penetrating power using Franz diffusion cell method, its stability, and its safety. Antioxidant activity of angkak extract was examined with DPPH method. Sonification method with surfactant RH40 and glycerin-cosurfactant (2:1) for a minute on frequency of 30% was used to create nanoemulsion of angkak extract. Characterization of extract angkak nanoemulsion included particle size, polydispersity index, and zeta potential. Angkak extract cream, nanoemulsion cream of angkak extract, and negative control cream were 3 kinds of preparation created from the cream, where all was further tested to find the stability. Cream safety test was conducted to animal with observation of the IIPR and IIO value. Cream penetration power test used Franz diffusion cell method. Result of this research showed that angkak extract has high antioxidant activity value, IC50 of 2.60 ppm. Chosen nanoemulsion of angkak extract has particle size of 151.60 nm, polydispersity index of 0.216, and zeta potential of 11.967 mV. Angkak extract cream and nanoemulsion cream of angkak extract safety test showed that the cream preparation is safe to use with IIPR value of 1.33 and IIO value of 0.00. Nanoemulsion cream of angkak extract has penetration power (Q) = 3,897 µg/cm2, higher than angkak extract cream with Q = 766.26 µg/cm2 on the 120th minute.
