Abstract
The enzyme amylase is an enzyme which has the capability of deciding bonds glycosides found in the polymer compounds, carbohydrates, starch molecules will be broken down by amylase on bonds of α-α-glycosides and 1,41,6-glycosides. The enzyme amylase can be produced by bacteria. The research of using waste coal mine water as a source of the enzyme amylase-producing bacteria by how to isolate, identify and characterize bacterial amilolitik for knowing his ability to produce amylase enzyme with clear zone measuring starch agar media. The results of this research get 5 isolates of bacteria with different characterization of bacterial isolates namely KPJ KPJ 1, 2, 3 and 4 KPJ KPJ has a spherical shape and KPJ 5 has irregular shape, elevation, the edges of the slick and white Lake dairy. Five isolates of bacteria is Gram positive, have a capsule and berspora.. Five isolates of bacteria have the ability to produce amylase enzyme to form clear zones on starch agar media. Clear zone measurement data contained on the highest-value 4 isolate 14,3 mm, then isolate the value 3 with 13,5 mm, with a value of 2 isolate 13,4 mm and on the value of the isolate 12,6 mm and the smallest is found in the clear zone isolate 1 with a value of 10,2 mm. Keywords: water, waste, coal, isolate, bacteria, enzymes, amylase.
