Abstract

Loratadin is an antihistaminic contains the BCS (Biopharmaceutics Classification System) class II with low solubility in water. Low solubility can be corrected by created ODT (Orally Disintegrating Tablets) through the formation of inclusion complexes. This study aims to determine the effect of formation of loratadine-β-cyclodextrin inclusion complex on loratadine solubility, the effect of the combination and proportion of Ac-Di-Sol with Crospovidon as superdisintegrant on disintegration time and dissolution of loratadine ODT. Inclusion complex formation is carried out using the method of kneading, loratadin: comparison of β-siklodekstrin ratio 1:2, characterized by FourierTransform Infrared (FTIR) and Differential Scanning Calorymetry (DSC). Variations in the combination of Ac-Di-Sol and Crospovidon with a ratio of 5%: 0%; 3,75%: 1,25%; 2,5%: 2,5%; 1,25%: 3,75%; 0%: 5%. The ODT tablets produced were evaluated with several parameters, namely weight uniformity test, hardness test, fragility test, wetting time test, in vitro and in vivo disintegration test, content uniformity test, taste response test and dissolution test. Data were analyzed by oneway ANOVA with 95% confidence level. The results showed that the loratadine-β-cyclodextrin inclusion complex could improve loratadine solubility. A large proportion of Ac-Di-Sol can slow down disintegration and dissolution times, Crospovidon at high concentrations will accelerate disintegration and dissolution times. The proportion of a combination of Ac-Di-Sol 1,25% and Crospovidon 3,75% results in rapid dissolution disintegration time.
