Abstract

Metformin hydrochloride is an effective antidiabetic drug for people with type 2 diabetes mellitus. Metformin hydrochloride is a class III Biopharmaceutical Classification System (BCS) drug with high solubility but low permeability. These drugs are available on the market in branded and generic preparations, so there may be differences in formulation and fabrication methods. This study aims to determine the physical quality and dissolution profile of innovative and generic metformin hydrochloride tablets. The test product used five 500 mg metformin hydrochloride tablet products, namely innovator drug (OI), 2 branded generic drugs (ODA and ODB), and two generic drugs with logo (OGC and OGD). Dissolution test using a type 2 (paddle) dissolution device with a speed of 50 rpm in 1000 mL dissolution medium. The dissolution medium used was phosphate buffer solution pH 6,8. Determination of dissolution rate using UV-Vis spectrophotometer at 233 wavelength. Result of research for physical quality test that is uniformity of measure, hardness, crushed time, and uniformity of weight that all product tablet of metformin hidroklorida have difference but still fulfill the requirement. The result of Q30 value indicates that all medicines fulfill the requirement that is solubility rate as much as 80% in 30 minutes. ODA products show that F1 value is less than 15 and F2 value more than 50 so it has dissolution profile which is similar to drug innovator.
