Abstract

Nifedipine is an antihypertensive calcium antagonists group. Nifedipine has a poor solubility in water. Nifedipine previously dispersed with PEG 6000 to improved solubility. Nifedipine preparations are made fast disintegrating tablet (FDT) is intended to provide rapid tablet disintegration time so as to provide rapid therapeutic effect. The aim of this research is to optimize the combination of AcDi-Sol and effervescent component as a disintegrnt of the physical properties of the tablet and release of the drug by the method of factorial design. The study was conducted using a factorial design with two factors. These two factors were Ac-Di-Sol and effervescent component. The concentration of Ac-Di-Sol used was 2% and 6%. The concentration of effervescent components used 3% and 12%. FDT nifedipine prepared by direct compression method. Tablet was evaluated by the physical properties of the tablet and the drug release. Optimum formula was determined on tablet properties hardness, friability, disintegrating, wetting time, Q1, and DE 30. Test perceptive sense also performed to determine the sweet taste in tablet FDT. Optimum formula was determined by counter plot superimposed on design expert software trial version 8.0.06. The results showed effervescent components provide reduction in disintegration time, drug release from tablets of nifedipine compared to Ac-Di-Sol and the tablets had a sweet taste. Based on the counter superimposed plot obtained optimum tablet formula with Ac-Di-Sol 6:00% and effervescent component 6:31%.
