Abstract

Ketoprofen is a propionate acid derivate use to treatment of non-steroid anti inflammatory drugs (NSAIDs) analgetic, antipirettan and anti-inflammatory. According to the Biopharmaceutics Classification System (BCS) ketoprofen is included in second classes drug which have low solubility and high permeability, so that ketoprofen is made in the preparation of orally disintegrating tablet (ODT), which is a solid dosage from with a suitable carrier is intended to disintegrate rapidly in the mouth upon contact with saliva in less than 60 secon. Using a combination of superdisintegrant crosscarmellose sodium and crospovidone. This research purposed to optimize the influence of combination crospovidone and crospovidone which have function as disintegrant, toward physical properties and drug release with factorial design method, which has four variations of a combination formula crosscarmellose sodium and crospovidone. The parameters are optimized hardness, friability, wetting time, disintegration time in-vivo, disintegrating time in-vitro, the amount of drug release of 1 minute (Q1), and the amount of drug release for 10 minutes (DE10). This study has shown that the components crospovidone give greater influence on the physical properties and the release of the drug, the optimum formula ketoprofen combination crosscarmellose sodium 5,19 mg and 10 mg crospovidone.
