Abstract

Theophylline and isoniazid is a drug that metabolized by the enzyme cytochrome P450. Theophylline given together with isoniazid can increase theophylline levels in blood plasma. The purpose of this research is to observe the effect of isoniazid to the theophylline phrmacikonetic profile in Wistar male white rats and how doses isoniazid gives changes significant theophylline pharmacokinetic profile. The study was using animals were divided into three groups with the total of the male rats are 9. Group I was given a single oral theophylline 2,34 mg/200grBW. Group II was given a theophylline oral together with isoniazid 5,4 mg/200grBW. Group III was given a single oral theophylline together with isoniazid 2,7 mg/200grBW. The serial blood was collected from orbitalis vein for 12 hours and were analyzed using HPLC mobile phase acetate buffer-acetonitril (93:7). The pharmacokinetic parameters of theophylline were calculated and were analyzed by One Way ANOVA with 95% degree of confidence. The result showed that added of isoniazid with doses 27 mg/kgBW do not give significant effect to theophylline pharmacokinetic profile in wistar male white rats.
