Abstract

Ranitidin HCL is a drug that it has low bioavailability and the best absorbtion in stomatch so that it can be formulated as gastroretentive floating tablet. Effervescent component, such as citric acid is factor that influencing floating ability and drug release. The aim of this research is determine the effect of citric acid to the floating lag time and drug release in ranitidin HCl floating tablet. The research used five formulas with content of citric acid 0 mg, 15 mg, 30 mg, 45 mg, and 60 mg. Tablet is made by wet granulation methode with average weight 400 mg and the hardness is controlled by 8 kg. floating test is done by inserting the tablet to the disolution medium (HCl 0,1 N pH 1,2) and the times were used the tablet to float is recorded. Dislution test used USP Aparatus II disolution test methode with HCl 0,1 N pH 1,2 as disolution medium and temperature was controlled in 370C until 6 hour. Sampling was taken at 15, 30, 60, 90, 120, 180, 240, 300, and 360 minutes. The concentration of ranitidin HCl was analysed by UV-Vis spectrophotometer with wavelength in 315 nm. The result of floating lag time test show that increasing of citric acid will be decreasing floating lag time. This is indicated by the formula that consist of the highest ccitric acid (formula V, 60 mg) has the most fast floating lag time (29,10 seconds) compare the other formulas. The result of drug release test show that increase of citric acid will be increase drug realease. This is indicated by the formula that consist the highest citric acid has the biggest drug who released. It is shown in value of DE360 (21,51%). This is bigger than all.
