Abstract
Kembang sepatu (Hibiscus rosa-sinensis L.) is a plant which use as drug diarrhea. Kembang sepatu leaves chemical constituents are flavonoid, alkaloid, tannin, saponin, sterol, and terpenoid. The purpose of the study was to determine the activity of n-hexane, ethyl acetate, and water fractions from methanol extract kembang sepatu leaves (Hibiscus rosa-sinensis L.) as antibacterial against E.coli ATCC 25922. Kembang sepatu leaves bark extraction was conducted by soxhletation method using methanol as solvent followed by fractionation with n-hexane, ethyl acetate, and water as solvents. Antibacterial activity were determined by diffusion and dilution method. Diffusion method was used to measure screening of antibacterial activity against bacteria colonies with concentrations used were 25%, 12,5%, and 6,25%. Dilution method was used to measure MIC and MKC conducted in serial dilutions of various concentrations 25%, 12,5%, 6,25%, 3,12%, 1,56%, 0,8%, 0,4%, 0,2%, 0,1%, 0,05%. Analysis of Varian to determine the most active fraction. Chemical content analysis conducted by TLC. The results showed that ethyl acetate fraction of methanol extract kembang sepatu (Hibiscus rosa-sinensis L.) leaves had the highest activity against E. coli ATCC 25922 compared to n-hexane and water fractions. Inhibition diameter of ethyl acetate fraction at concentration 25%, 12,5%, and 6,25% against E.coli ATCC 25922 respectively 20,67 mm, 18,33 mm, 16,17 mm and had Minimum Killing Concentration of 6,25%. The presence of flavonoid and sterol in ethyl acetate fraction were detected in the preliminary phytochemical test.
