Abstract

Theophylline is a bronchial smooth muscle relaxant which has a history of use for treating bronchial asthma and other respiratory diseases. Theophylline undergo biotransformation in the liver by the enzyme cytochrome P-450 (CYP) and only 10% excreted through the kidneys in unchanged enzyme responsible is CYP 1A2, 2D6, 3A3 / 4 and 2E1. Pineapple is a fruit that is often consumed by people. Pineapple contains quercetin which may interfere with the metabolism of theophylline on CYP CYP 1A2, 2D6, 3A3 / 4 and 2E1. This study aims to determine the pharmacokinetic profile of theophylline when administered in conjunction with pineapple juice. This study tests was using 9 rabbits divided into 3 groups. The first group, the theophylline dose group controls 7 mg / KgBW, Group II, theophylline 7 mg / KgBW and pineapple juice 7.02 g / KgBW, and Group III theophylline dose of 7 mg / KgBW with pineapple juice double dose of 14.04 g / KgBW. Analysis of plasma levels using a C-18 HPLC column and mobile phase may acetate and acetonitrile 93: 7. Snippets of blood taken at minute 15, 30, 45, 60, 120, 180, 240, 300, 360, 420, 480, 540. The data were analyzed by ANOVA. The results obtained, a double dose of pineapple juice can increase the AUC of 53.21%, decreasing by 32.01% CLT, and lower K 21.75%. The third parameter indicates that quercetin found in pineapples can interfere with the metabolism of theophylline when administered at a dose of pineapple 14.04 g / kg rabbit. .
