Abstract

Breast cancer is the most cancer in Indonesia. Carissa carandas L. is a most plant of Indonesia which si widely used traditionally as a medical plant. Carissa carandas fruits contain flavonoids, alkaloids, tannins, triterpene and steroids that has potent activity as anticancer. The aim of this study was to find out cytotoxic of chloroform fraction of Carissa carandas fruit using T47D cells and vero cells. This research was conducted using MMT method. The principle of this method is the ability of reductase mitochondria that change the methylthiazol tetrazolium to purple formazan crystal. This research was performed using variation concentrations: 500; 250; 125; 62,5; 31,25; 15,625; 7,81 mg/mL. The value was read using ELISA reader absorbance wavelength of 595 nm then converted in percentage viability of living cells. IC50 value was obtained from regression plotting between log concentration and percentage viability. This research was obtained chloroform fraction of Carissa carandas contain flavonoids, triterpene and steroids. The IC50 value of chloroform fraction of Carissa carandas fruit on breast cancer cells T47D and vero cells is 3.411,9 µg/ml and 879.022,516 µg/ml with selectivity index of 257,6. The result of this study concluded that the chloroform fraction of the metanol extract of Carissa carandas fruit have low toxicity effect against breast cancer cells T47D and high selectivity index.
