Abstract

Hibiscus plant (Hibiscus rosa-sinensis L.) is used as a traditional medicine among white discharge and canker sores. Hibiscus leaves contains flavonoid (quercetin), saponin, tannin, terpenoid, alkaloid and sterol. This study aims to determine the antifungal activity of the fraction of n-hexane, ethyl acetate and water from hibiscus leaves methanolic extract against Candida albicans ATCC 10231. Hibiscus leaves powder was extracted by soxhletation method by methanol, and then it was fractionated by solvent n-hexane, ethyl acetate and water wich have different polarity. Antifungal activity test was performed using diffusion and dilution methods. The concentration used in the diffusion method was 50%, 25% and 12,5% aimed to determine the most active fraction.The most active fraction is continued dilution test to determine the MIC and MBC with concentration of 50%, 25%, 12.5%, 6.25%, 3.125%, 1.563%, 0.781%, 0.391%, 0.196%, 0.098%.Statistical analysis using oneway ANOVA to determine whether there is a significant difference between the test preparation. The results shows that all the fractions and extracts has antifungal activity. Ethyl acetat fraction is most active fraction with 25,3 mm diameter inhibition at concentrations of 50%, 25% concentration of 26,3 mm and 20,7 mm concentration of 12,5%. Dilution test results ethyl acetate fraction showed antifungal activity with MBC 12,5%. Analysis contents of the compounds in ethyl acetate fraction used TLC showed flavonoids and sterols.
