Abstract

Cetirizine HCl (CTZ) is an antihistamine drug that works as an H1 receptor antagonist, many formulated as a tablet dosage form. The tablet is a pharmaceutical preparation of practical use yet dysphagia or difficulty swallowing would be difficult for the patient then made preparations fast disintegrating tablet (FDT) Cetirizine HCl. Filler-binder in the preparation of the FDT can affect the physical quality of the tablet so it needs to be optimized. This study was conducted to determine the effect of the combination of Starch 1500® and manitol on the quality of the physical and the release of active substances as well as to determine the most optimum formula. This study uses simplex lattice design with two factors namely Starch 1500® and manitol in formula 1; formula 2; Formula 3 in sequence 100%: 0%; 50%: 50%; 0%: 100%. Tableting was conducted using a direct compression then tested the physical quality dust of tablet and tablet physical quality test. Determining the optimum formula based counterplot obtained from Design Expert 8.0.7.1 trial version with parameter critical point hardness, friability, disintegration time in vitro, Q5 and DE30. The results showed that Starch 1500® greater the increase fragility, Q5 and DE30 while the manitol larger to increase the hardness and in vitro disintegration time. Starch 1500® 32.05% (50.64 mg) and Manitol 67.95% (107.36 mg) are proportion of produced a optimum formula with physical quality and the most excellent drug release.
