Abstract
Propranolol HCl is a drug used in the treatment of hypertension, angina pectoris, and other cardiovascular problems that work against β-adrenergic receptors by inhibiting non-selective adrenergic stimulant response. Propranolol HCl is stable in acidic, well absorbed in the stomach, and has a short half-life of about 2-6 hours, so it can be developed for propranolol HCl sustained release dosage retained in the stomach. This study aims to optimize and evaluate ethyl cellulose and natrium bicarbonate ablity to float and drug release in tablet gastroretentive on propranolol HCl with a floating system. This study was conducted with a variation of the four formulas with a ratio of ethyl cellulose and natrium bicarbonate at a different F1 (5%: 8%), Fa (20%: 8%), Fb (5%: 15%) (20%: 15% ). Test drug release is done with the model paddle USP apparatus II model, with a stirring speed of 50 rpm at 37 ± 0.5 ° C using a mediumHCl pH 1.2 for 6 hours. Optimization is done against hardness, floating lg time, swelling index, Q60, Q180, and Q300 uses Expert® Design program. The results showed that the ethyl cellulose has a dominant role to increase hardness, swelling index, and drug release. Natrium bicarbonate can reduce floating lag time. Factorial design examination found the optimum formula of ethyl cellulose 80 mg and 59 mg of sodium bicarbonate. Keywords: propranolol HCl, ethyl cellulose, natrium bicarbonate, factorial design
