Abstract

Black pepper seeds (Piper nigrumL.) and fruit of mahkota dewa (Phaleria macrocarpa (Scheff.) Boerl) on previous research has antibacterial activity against Escherichia coli. The purpose of this study to determine the activity of the combination of black pepper seed extract and mahkota dewa in various combinations. Black pepper seeds (Piper nigrumL.) and fruit of mahkota dewa (Phaleria macrocarpa (Scheff.) Boerl) powdered and extracted by maceration method using 70% ethanol. The booth of extract are combined with a ratio (1:1), (1:2), and (2:1), made serial dilution of 100 mg/ml; 50 mg/ml; 25 mg/ml; 12.5 mg/ml; 6.25 mg/ml; 3.125 mg/ml; 1.56 mg/ml; 0.781 mg/ml; 0.3905 mg/ml, tested against Escherichia coli ATCC 25922 in dilution. Antibacterial test results showed that the combination of 70% ethanol extract of black pepper seeds and fruit of mahkota dewa have antibacterial activity on a comparison of the combination (1:1) and (2:1) with Minimum Bactericidal Concentration (MBC) respectively 100 mg/ml and 50 mg/ml. Comparison (1:2) had no antibacterial activity at a concentration of 100 mg/ml. Comparison of the most effective combinations are (2:1), this is possible because pepper has a class of alkaloids (piperine).
