Abstract

Leaves Carandas (Carissa carandas L.) is one of the plants that can be utilized as a traditional medicine. The genus Carissa has activity as anticancer, including the species of Carissa cipanarum which has been used as a traditional medicine in India as an anticancer (Fatimah 2013). This study aims to determine the cytotoxic activity of extract and water fraction of carandas leaf by in vitro as well as determine IC50 value of methanol extract and water fraction. Test extract as much as 10 mg subsequently dissolved in 50 mL of DMSO (Dimethil sulfoxide) in Eppendorf for each test extract and stored as stock solution. The well was added 100 mL of test solution i.e. methanol extract in medium of each well in order to obtain final concentration of samples with variations in certain concentrations (500; 250; 125; 62,5; 31,25; 15,625; 7,81) µg/mL per/well, as a control used cell without addition of test solution. Analysis data by calculating percentage of death value which obtained from 100% - % living cell, then performed data analysis using linear regression by changing mortality percentage into probit to determine IC50 value. Based on the IC50 obtained from the results of this study concluded that the water fraction of the methanol extract of leaves carandas have anticancer activity against HeLa cells were rather low, with IC50 value of 32838 mg/mL.
