Abstract

Multiple sclerosis (MS) is a chronic inflammatory disease of the central nervous system (CNS) associated with demyelination, axonal weakness or atrophy of the brain. The content of steroids in takokak fruit (Solanum torvum Swartz), is expected to have an effect in reducing the number miroglia immunosuppressants in MS. This study aims to determine the effect of n-hexane fraction takokak fruit against histopathological and number of brain microglia and effective dose in mice models of MS. A total of 35 rats were divided into seven groups, each group consisting of 5 mice. 1 as the normal control group were not given treatment, 2 as a negative control group were given CMC Na 0,5% and 3 as the positive control group were given methyl prednisolone 45 mg / kg BW. Groups 4, 5, 6, 7 were given fraction of n-hexane with successive doses; 70 mg / kg, 140 mg / kg, 280 mg / kg, 560 mg / kg. On day 0, group 2-7were induced with 0.2 ml ensefalitogen sc and 0.5 ml sc Pentabio® vaccine. On2ndday,groupsof2-7 wereinducedwith0.5mlPentabio®vaccinesc.Groups2-7weregiven thetestpreparationfromdayto14-30.Eachday,theratswereobserved the neurologic symptoms. On day 31, the mice were sacrificed and brain histopathology test. Data were analyzed using one way ANOVA. The results showed that n-hexane fraction takokak fruit affect the histopathological picture and the number of brain microglia in the mice model of MS with the effective dose of 70mg/kg BW.
