Abstract

The amount of cervix cancer is a problem that need consideration. Carissa carandas L that has traditionally been used as herbal medicine. Carissa carandas L leaf contains terpenoid, saponin, tannin, alkaloid, flavonoid and steroid that has potent activity as anticancer. The aim of this study was to find out the cytotoxic activity of chloroform fraction of methanol extract of Carissa carandas L leaf by in-vitro study toward HeLa cell. This research was conducted using MMT method with RPMI medium. The principle of this method is the ability of reductase mitochondria dehydrogenase on active cell that change the tetrazolium salt to formazan crystal (purple color) that could be adsorbed by ELISA reader at wavelength 595 nm. This research was performed using seven concentrations i.e. 500; 250; 125; 62,5; 31,25; 15,625; 7,81 mg/mL. The absorbance was conversed to percentage of cell viability. IC50 value was obtained from regression plotting between log concentration and viability Chloroform fraction contains terpenoid, alkaloid, flavonoid and steroid. This research was obtained IC50 value of chloroform fraction of Carissa carandas L leaf of 76 µg/mL. It can be concluded that chloroform fraction of Carissa carandas L leaf has toxicity effect toward cervix cancer. Keywords: HeLa cell, Carissa carandas L, Chloroform fraction, IC50
