Abstract
Isoniazid is an izonicotinic acid derivate, it have been used as tuberculosis treatment. Inactivation isoniazid in liver trough acetylation process form acetyl isoniazid followed by hydrolysis reaction become free acetyl hydrazin is changed as toxic metabolic by cytochrome P450 enzyme. Curcuma® tablet is a product from extract of Curcuma xanthoriza that consist of curcumin as active ingredient. Curcumin have been reported able to inhibit the activity of cytochrome P450 1A1/1A2 and 3A4. The presence of Curcuma tablet in isoniazid treatment is expected to reduce the hepatotoxic effect of isoniazid. This research aimed to determine the influence of Curcuma tablet on pharmacokinetic profiles of isoniazid orally given to wistar rat. This research was divided in three groups i.e. control group was treated with isoniazid 5 mg/kg body weight, treatment group was treated with isoniazid 5 mg/kg body weight and curcuma tablet 200 mg/70kg body weight in 1 hour before isoniazid administration and during 7 days, respectively. Blood was taken through opthalmicae vein with sampling interval 0.5, 1, 1.5, 2, 3 and 4 hours. Isoniazid assay in blood was analyzed using high performance liquid chromatography (HPLC). The results showed that Curcuma tablet statistically (p>0.05) not affected the pharmacokinetic profiles of isoniazid using several parameters i.e. time of maximum concentration, area under curve, distribution volume, elimination rate and absorption rate.
