Abstract

Lactose is a disaccharide consisting of D-glucose and D-galactose and called chemically 4-0-β-D-galaktopiranosil-D-glucopyranose. Lactose has been knowed as milk sugar because it is only found in milk. This study aims to know the content of lactose that found in low-fat milk in the UV-Vis spectrophotometry. This study uses five samples of low-fat milk in the box packaging. The first phase was conducted with a qualitative test on three test samples using a chemical reaction, that is molish test, barfoed test and benedict test then continued using UV-Vis spectrophotometry. Determination of lactose content in the sample is to find the maximum wavelength and the operating time. The wavelength that obtained is 749.5 nm and the operating time 10 – 23 minutes. The conditions used to read the calibration curve and the assay sample. Based on the results of research obtained lactose content in the low-fat milk packaging for samples A 3.38%; B 6.99%; C 13.13%; D 7.87%; and E 7.41%. Compared with the average composition of low-fat milk according to Buckle 1987 contained 5% lactose content, only one of the fifth milk sample is a sample B that close to these requirements.
