Abstract

Domperidone is classified under class-II (BCS). This poor solubility is an appropriate alternative to made orally disintegrating tablet domperidone formulations so increase covenient of use in pediatric and geriatric patients with give a good teste and faster disintegration in mouth. This study was aimed to find out the optimal concentration and effect of combination superdisintegrant sodium starch glycolate and croscarmellose sodium to physical properties and disolution rate of ODT domperidone. The simplex lattice design used to find effect and interaction of tablet physical properties and disintegration rate of the domperidone tablet with critical points, such as flow rete, water absorption, disintegrating time, wetting time, and disolution use combination of sodium starch glycolate and croscarmellose sodium with ratio 75:25; 50:50; 25:75, weight of tablet 200 mg wiht direct compression methode. After the optimum formula is obtained then be compared the experimental result and the prediction result then analized with one simplex t-test. The optimum formula is consentration 22,5 mg and 7,5 mg (sodium starch glycolate:croscarmellose sodium) with one simplex t-test result ρ>0,05 so that there was no significant defference between the experimental result and the prediction result. The addition of croscarmellose sodium is relatvely low compared the sodium starch glycolate will give the faster disintegration time, wetting time, and disolution and show decrease of hardness and friability of the tablet.
