Abstract

This study used natural ingredients i.e. black pepper seeds (Piper nigrum L.) which was efficacious as an antioxidant as well as an antihypertensive. The ethanol extract of black pepper seeds are known to have high antioxidant activity. The purpose of this study was to determine the antioxidant activity of extract, fractions and sub-fractions of black pepper seeds and know the class of compounds that play a role in antioxidant activity. Extract of black pepper seeds was obteined by maceration, was fractionated with the solvents of n-hexane, ethyl acetate and water. The fraction with the highest antioxidant activity was continued to be sub-fractionated by vacuum liquid chromatography (VLC). Extract, fractions and sub-fractions was tested their antioxidant activity by 1,1-diphenyl-2-picrylhydrazyl (DPPH) method and quercetin was used as a standart. Sub-fraction with the highest antioxidant activity was identified by Thin Layer Chromatography (TLC). The antioxidant test results showed that ethanol extract, n-heksan fraction, ethyl acetate fraction and water fraction of black pepper seeds had IC50 of 374,87±0,98, 1426,83±4,43, 275,48±1,11 and 992,49±3,24 ppm respectively. Antioxidant activity test of ethyl acetate sub-fraction showed that F sub-fraction of black pepper seeds had smallest IC50 value of 56,21±0,41 ppm.
