Abstract

Pepper leaves (Piper nigrum L.) contain flavonoid, saponin, volatil oil and alkaloid. The purpose of this study was to determine the activity of n-hexane fraction, ethyl acetate fraction, the fraction of water and ethanol extracts of leaves of pepper (Piper nigrum L.) as an antibacterial against Escherichia coli ATCC 25922. Pepper leaves powder macerated with 96% ethanol, and then fractionated using n-hexane, ethyl acetate, and water solvents. Ethanol extract, n-hexane, ethyl acetate, and water fractions were tested antibacterial activity using dilution method with concentrations of 50.0%; 25%; 12.5%; 6.2%; 3.1%; 1.5%; 0.7%; 0.3%; 0.1%; 0.09% of the bacteria Escherichia coli ATCC 25922. The results showed that the fraction of n-hexane, ethyl acetate, water, and pepper leaf extract has antibacterial activity against Escherichia coli with a minimum killing concentrations levels - contribute 50 %; 12.5 %; 50 %; and 50 %. Ethyl acetate fraction of leaf pepper has antibacterial activity compared to the most active fraction of n - hexane, water and extract.
