Abstract
In the pathological state such as diabetes, increased oxidative stress in the body will cause decrease in endogenous antioxidant activity in the body so that the body do not able detoxify free radical and prevent cell damage. One source of natural antioxidant as antidiabetic is black cumin seeds (Nigella sativa L.). The purpose of this study was to determine antihyperglycemic activity of black cumin seeds ethanol extract and antioxidant activity of SOD and GPX enzymes and decreased MDA level. This study was an laboratory experimental research using a post-test only group design. The subjects of this study were 25 male Wistar rats which conditioned type 2 diabetes by alloxan induction. Rats were grouped into 5 groups: group I normal, group II negative control alloxan 150 mg/kg BW, group III positive control glibenclamide 0,45 mg/kg BW, group IV dose of ethanol extract of black cumin seed 125 mg/kg BW and group V dose of ethanol extract of black cumin seed 250 mg/kg BW. The results showed that ethanol extract of black cumin seed dose of 125 and 250 mg/kg BW could decreased blood glucose levels of rats and MDA and increased the activity of SOD and GPx. The ethanol extract of black cumin seed doses of 125 and 250 mg/kg BW could regenerate pancreatic islet cells damage induced alloxan. The most effective dose in decreased blood glucose levels and MDA and increased the activity of SOD and GPx was dose of 250 mg/kg BW. Dose of 250 mg/kg BW was also the most effective in regenerating pancreatic cells in this study. Keywords : Nigella sativa L, antihyperglycemic, antioxidant, pancreas regeneration.
