Abstract

Antioxidants are compounds important in maintaining a healthy body because it functions as a catcher of free radicals. Breadnut (Artocarpus communis J. R. & G. Forst.) leaves contains flavonoids, saponins, tannins, and polyphenols. The purpose of this study was to determine the antioxidant activity of a fraction of n-hexane, ethyl acetate fraction, the fraction of water, and ethanolic leaves extract against DPPH radical breadnut using IC50 parameters and determine the fraction of the ethanolic extract of the leaves breadnut which has the highest activity. Breadnut leaves powder was macerated using 70% ethanol then the ethanol extract was fractionated using n-hexane, ethyl acetate, and water. Compounds of extracts and fractions were identified by TLC. Fractions and extracts were tested against radical DPPH antioxidant activity using a spectrophotometer and used rutin as positive control. After testing the antioxidant the value of IC50 was determined. The results showed that IC50 value of n-hexane fraction 85,97 ± 3,57 ppm, ethyl acetate fraction 47,59 ± 1,02 ppm, the water fraction 51,34 ± 1,09 ppm, and extract ethanolic 71,19 ± 3,71 ppm. Rutin as the comparison had IC50 of 5,36 ± 0,10 ppm. Ethyl acetate fraction and water fraction had the highest antioxidant activity. Keywords: Artocarpus communis J. R. & G.
