Abstract

Diabetes mellitus (DM) is a metabolic disorder of carbohydrates, lipid and protein in which blood glucose can not be used well that, cause the hyperglycemic state. The brotowali (Tinospora crispa L. Miers) stem bark was reported to have activity hypoglycemic and antioxidant (SOD,GPx and MDA) and regeration ability on the pancreas in alloxan induced rats. This study was an laboratory experimental research using a post-test-only-groupdesign. The subject of this study were 30 male. Diabetic rat (group II-VI) was induced using alloxan. The rats were divided into 6 groups : group I (normal control), group II negative (control Na-CMC), group III positive control (glibenclamide 0,45 mg/kgbw), group IV water fraction of brotowali dose of 30 mg/kgbw, group V water fraction of brotowali dose of 60 mg/kgbw, group VI water fraction of brotowali dose of 120 mg/kgbw (Each group were tested a SOD activity, GPx and MDA continued by test histopatologi) in the study, blood glucose levels, SOD, GPx and MDA activities were also evaluated and the pancreatic cells were observed Histopb/b /bathologically. This study showed that water fraction of stem bark ethanol extract brotowali (Tinospora crispa L. Miers) at dose 30, 60, 120 mg/kgbw has activity of lowering blood glucose levels significantly in comparison to that negative control (p<0,05). A dose which most effective in lowering blood glucose levels, increase of SOD activity, GPx and lowering levels of MDA as well as be able to repair damaged cells in pancreatic island was 120 mg/kgbw. Keywords: Tinospora crispa L. Miers, water fraction, alloxan, antioxidan, pancreas regeneration.
