Abstract

Syzgium cumini and Tinospora crispa are medicinal plants that can be used in the treatment of diabetes melitus. The study aimed to determine the activity of combination of ethyl acetate fraction of Syzigium cumini L. and water fraction of Tinospora crispa on blood glucose level and translocation of GLUT-4 in insulin resistant rats. The study was conducted using Wistar rat (Rattus norvegicus) divided into 6 groups. Group 1: normal control, group 2: negative control (Na CMC), group 3: positive control (metformin 45 mg/kg BW), combination groups ethyl acetate fraction of Syzygium cumini cortex : water fraction Tinospora crispa cortex respectively from group 4 to group 6: 25:86,25; 50:57,5 dan 75: 28,75 mg/kg BW. Insulin resistant was induced by administration of 1.8 g fructose / kg body weight and fat diet for 45 days.Assay of glucose using a GODPAP method. Observation of GLUT-4 expression in muscle tissue by using immunohistochemical techniques. Insulin resistant rats was tested by using two parameters: (1) test of blood glucose level preprandial and postprandial (2) observations of GLUT-4 protein expression in muscle tissue. Results of antidiabetic activity test showed that combination group of ethyl acetat fraction Syzygium cumini cortex 25 mg/Kg BW and water fraction Tinospora crispa cortex 86,25 mg/kg BW had the most optimal activity to percentage of decrease in blood glucose levels by 48,74 % and to decrease of GLUT-4 expression in soleus muscle as 80,94 % to conditioned DM type 2 insulin resistant rats.
