Abstract

Mangosteen (Garcinia mangostana L) a fruit that a antidiabetic activity. mangosteen rind contains a main active compound flavonoid and xantone a antidiabetic properti. Type 2 diabetes mellitus type 2 can be caused by insulin deficiency and insulin resistance. This study aims to determine the activity of ethanolic extract of mangosteen rind in decrease blood sugar levels and increas GLUT-4 protein activity in decresing insulin resistance wistar rats Method used in the study was ethanolic extraction of mangosteen rind. The animals were used divided into 6 groups. Group 1: normal control, group 2: negative groups; Group 3: positive control (metformin 45 mg / kg), group 4: the ethanol extract dose of 50 mg / kg, group 5: ethanol extract dose of 100 mg / kg, group 6: ethanol extract dose of 200 mg / kg. Animal tests ( Group 2-6) were given fructose and high fat diet (HFD) to induce insulin resistance. Blood glucose levels were observed at day 0, day 15 and 30 as pre-treatment, and after treatment on day 37 (T2) and on day 45 (T3). Insulin resistance assay was at done fasting blood glucose levels and observation of GLUT-4 protein expression in the muscle tissue. This study used a one-way ANOVA analysis with a level of 95%. Results showed that group dose of ethanol extract mangosteen pericarp 100 mg / kg was the most optimal dose in lowering blood glucose levels and increase the value of GLUT-4 protein translocation.
