Abstract

Multiple Sclerosis (MS) is an autoimmune disease because of the damage in myelin sheath that mediated by T cell. The steroid content in takokak fruit (Solanum torvum Swartz) is expected to give an immunosuppressant effect to suppress phagocytosis macrophage in MS. This research aims to know the effect of n-heksan fraction of takokak fruit and the effective dose for the reduction of phagocytosis macrophage of rat of multiple sclerosis model. There are 35 rats being divided in 7 groups. Group 1 as the normal control was not treated. Group 2 as negative control were given Na CMC 0,5 %. Group 3 as positive control were given methylprednisolone 45 mg/kgBW. Groups 4,5,6, and 7 are given n-heksan fraction with consecutive doses: 70 mg/kg BW, 140 mg/kg BW, 280 mg/kgBW, and 560 mg/kgBW. On the day zero, the groups 2-7 are induced with encephalitogen 0,2 ml sc and Pentabio® vaccine 0,5 ml sc. On the second day, the rats of groups 2-7 are induced again with Pentabio® vaccine 0,5 ml sc. Test material is ad administered orally from day fourteenth to thirtieth. Each day, the rats were weighed and observed the neurologic symptoms. On the day thirty first, the rats are dissected for the phagocytosis macrophage test. The result were analyzed with one way ANOVA. The research results shows that the n-heksan fraction of takokak fruit is able to suppress the MS symptom wich shown by the reduction of the average score of neurologi, the reduction of the total sick days of rat , and has an effect to reduce the activity and capacity of phagocytosis macrophage of rat of MS model with the effective dose of 280 mg/kg BW of rat.
