Abstract

Rifampicin is a antituberculosis (anti TB) drug that degrades in stomach and its decomposition is affected by the presence of isoniazid in stomach after consumption. Curcuma® tablet contains curcumin that has been used as hepatoprotective and support treatment for TB, although the pharmaceutical interactions that might occur has not been studied. The aim of this study was to find out the effect of Curcuma® on concentration of rifampicin, isoniazid and their mixture by in-vitro study using high performance liquid chromatography (HPLC). This study consisted of six groups, three groups of control and 3 test groups. A control group was without Curcuma® and the test group was by giving Curcuma® performed in vitro. The assay of rifampicin and isoniazid were determined by the HPLC method using C-18 column. HPLC condition used the mobile phase of methanol: water (60:40), flow rate of 1.0 ml/min and wavelength at 236 nm. Results of in vitro studies showed that levels of isoniazid increased on addition of rifampicin, but after being given Curcuma® isoniazid levels not increased. Rifampicin levels decreased on the addition of isoniazid, whereas after Curcuma® added rifampicin levels increased. Increased levels of rifampicin showed that Curcuma® give effect to rifampicin is by increasing the levels of rifampicin, and Curcuma® did not affect the levels of isoniazid.
