Abstract

Meloxicam is a selective inhibitor of cyclooxygenase 2 enzyme that been used as rheumatoid arthritis treatment and has low solubility. Inclusion complex meloxicam in β- cyclodextrin be used to enhance solubility and taste masking. Formulation in fast disintegrating tablet dosage form can enhance patient compliance. This research aimed to determine the influence of Primellose® and Kollidone® CL on physical properties and drug release of FDT meloxicam/β-cyclodextrin complex. The inclusion complex was prepared by kneading method using 1:2 drug to β- cyclodextrin molar ratio and characterized using thermal analysis (difference scanning calorimetric; DSC). The tablet was prepared by direct compression method with several variation concentrations i.e. 1:1. 4:1, 1:8 and 4:8 Primellose® to Kollidone® CL ratios. Hardness, friability, wetting time, in vitro and in vivo disintegration time and dissolution as dependent variables. Data were analyzed statistically using analysis of variance (ANOVA, p-value set 0.05). The DSC thermogram showed that meloxicam loaded into β-cyclodextrin cavity due to disappearance of meloxicam melting point. Primellose® affected on an increase of hardness, in vitro, wetting time and a decrease of friability mean while Kollidone® CL affected on an increase of friability and a decrease of in vitro, wetting time. Primellose® of 1% and Kollidone® CL of 8% produced tablet less friable and rapid disintegration time.
