Abstract
Salbutamol sulphate is one of the widely used drug for the treatment of bronchial asthma, chronic bronchitis, and emphysema. Salbutamol has short half-life about 2-4 hours, with small bioavailability. Salbutamol sulphate stable at acidic condition and degradation in colon, so that should be developed in a sustained release dosage form are retained in stomach. This research purposed to optimize and evaluate the influence of HPMC K15M and effervescent components on floatation behavior and drug release of gastrorententive salbutamol sulphate tablet with floating system. This research was conducted on three formulas with HPMC K15M and effervescent component of different, ie F1 (60%:10%), F2 (55%:15%), and F3 (60%:20%). Dissolution test conducted using apparatus model II USP paddle model with 50 rpm whirling on 37 + 0,5oC using HCl 0,1 N during 6 hours. Optimation to analyze floating lag time, Q60, Q360, and DE360 by Design Expert® program. The result show effervescent component has dominant role on floating lag time. Interaction between two components increase floatation behavior. HPMC K15M can decrease drug release. Analyze using simplex lattice design show the optimum area of HPMC K15M is 100-120 and effervescent component is 20-40.
