Abstract

Pletekan Plant (Ruellia Turosa L.) is one of the medicinal plants used to treat diabetes mellitus. Obesity can cause insulin resistance and is one of the risk factors of diabetes mellitus. This study aims to determine the antidiabetic effects of leaf extract pletekan combination with metformin in mice induced insulin resistance with obesity. Test animals were divided into 6 group. I as the normal control group was given normal feed (normal diet), group II-VI given HFD feeding for 4 weeks. Body weight of mice were weighed every single week. Insulin resistance was tested by injecting insulin with insulin tes tolerancep 0.75 U/kg body weight (b) intraperitoneally. Test preparation is administered for 9 days, blood glucose levels were checked on day 5 and 9 after administration of the test preparation. I as a normal control group (distilled water), group II as a control group metformin (2.6 mg/20 g bb mencit), group III as the combination group pletekan leaf extract (15 mg/20 g bb mice), group IV as a group combined ethanol extracts 70% leaf pletekan: metformin 75% : 25% (11.25 mm mg/20g mice: 0.32 mm mg/20g mice), group V as a combined group of 70% ethanol extract of leaves pletekan: metformin 50%: 50% (7.5 mg/20 g bb mice: 0,65 mg/20 g bb mice), and group VI as Extract the combination group leaves pletekan: metformin 25%: 75% (3.75 mg/20 g bb mice: 0.97 mg / 20 g bb mice). Statistical data obtained by one way ANOVA (p <0.05). The results showed that all combinations of ethanol extract of leaves pletekan and metformin may lower blood glucose levels comparable to metformin in mice induced insulin resistance with obesity. Diabetes therapy with a combination of ethanol extract and metformin is recommended for people with diabetes mellitus because metformin dose can be lowered so that the side effects of metformin can be reduced.
