Abstract

Stevia rebaudiana Bertonii M. is one of the medicinal plant in Indonesia. Stevia plant beneficial in decreasing blood sugar level and blood pressure, antimicrobial, oraltonic and anticancer. The aim of this study was to know the ability of plant – growth regulator NAA and BAP in inducing stevia leaves callus, stimulating steviosida formation, to know concentration of plant – growth regulator NAA and BAP that can increase the concentration of stevioside and to know the concentration of stevioside in stevia leafs callus. This study used plant tissue culture method with New Phaleonopsis (NP) medium added with growth regulators NAA and BAP dosage 0 ppm : 1 ppm; 0,25 ppm: 0,75 ppm; 0,5 ppm: 0,5 ppm; 0,75 ppm: 0,25 ppm; 1 ppm: 0 ppm. Callus evaluation was conducted on the percentage of successful callus explants formed, time induction, and average callus weight. Analysis qualitative and quantitative content of stevioside was determined by dencitrometry TLC with mobile phase chloroform : methanol : water (10:15:2), and stationary phase silica F254. The results of the study showed that variations in the concentration of growth regulators NAA and BAP were able to induce callus growth of stevia leaf with the largest percentage of success on the addition of NAA and BAP 0.5 ppm 0.5 ppm is equal to 78.95%. Average weight is 1.2190 greatest callus on NAA addition of 0.5 ppm and 0.5 ppm BAP. The addition of plant growth regulators NAA and BAP are not able to increase the formation of steviosida. Steviosida greatest levels of NAA in the addition of 0 ppm and 1 ppm BAP at 0.043%. Keyword : Stevia leaf callus, New Phaleonopsis (NP), plant growth regulators NAA and BAP, stevioside
