Abstract

Meloxicam is a non-steroidal anti-inflammatory drugs for rheumatoid arthritis and osteoarthritis disease that requires rapid action. Meloxicam formulations in the preparation of fast-disintegrating tablets meloxicam in β-cyclodextrin inclusion complex is an appropriate alternative to increase comfort with a good sense and quickly disintegrate in the mouth. This study was purposed to optimize and determine the effect of Primellose® and Primojel® on the physical properties and drug release of FDT meloxicam/β-cyclodextrin complex. Inclusion complex of meloxicam in the β-cyclodextrin was prepared using spray drying method in 1:2 drug to β-cyclodextrin molar ratio. A 22 factorial design method was applied to optimize and assess the influence of Primellose® and Primojel® as independent variables. The optimum area defined by superimposed contour plot of wetting time, disintegration time (in vivo and in vitro), friability, the amount of drug released at 1 minute and 10 minutes and 10 minutes to the dissolution efficiency using Design Expert software. The results showed that increasing the time factors of Primojel® improve wetting and disintegration time (in vivo and in vitro) and lower brittleness compared with Primellose®. The interaction of these two factors affect the increased friability, wetting time, and the disintegration time in vivo, as well as lowering the in vivo disintegration time and reduces the cumulative amount of drug released in the early minutes. Based on superimposed contour plots obtained optimum formula Primellose® factor 2 mg and 13.82 mg Primojel®.
