Abstract
Theophylline has a relatively short half-life (8,1 jam) with a narrow therapeutic window is 10-20 µg/mL. sustained release formulation can produce more uniform serum concentrations with less fluctuation in peak trough level. The physical properties of tablet mass and the release profile of drug from hydrophilic matrices are influecend by properties of matrix components, i.e HPMC (gelling agent), Xanthan gum (free flowing). The research was done with simplex lattice design (SLD) by using 2 component, i.e HPMC K4M (A), Xanthan gum (B). three formula were obtained that are F1 (100% A : 50 % B), F2 (50% A : 50% B) F3 (50% A : 100% B). the optimization parameters of theophylline sustained-release were flow rate of the tablet mass, the dissolution efficiency of the tablet mass, and the release rate of theophylline. Based on SLD model ; equation, normal plot of residual. By wich the optimum formula was determined. The results of the study that combined theophylline matrix material HPMC K4M and Xanthan Gum can affect the physical properties of the tablet, as well as deliverance other men. The proportion who produce the physical properties of tablets and maximum release is a formula containing 15% HPMC K4M and Xanthan gum 30% of the drug.
