Abstract

Diabetes is now regarded as heterogenous group of diseases characterized by a state of cronic hyperglicemia, which causes a number of secondary complication. Complication is related to diabetes are associated with oxidative stress induced by generation of free radical. Antioxidan this play an important role to protect the human body against damage caused by reactive oxygen spesies and beneficial effect agains the diabetes complication. Bitter melon fruit( Momordica charantia L.) is one of the frequenly used as antidiabetic herbs in Indonesia. The aim of study is to evaluate the antihyperglycemia, antioxidant protective effect of Momordica charantia L. extract ethanol (MCE) in pancreas of aloxan induced diabetic rats. In this study, antihyperglycemia, antioxidant protective effect of MCE were evaluated using fasting blood glucose, enzim antioxidant (SOD, GPx, MDA) concentration and histophatology of pancreas diabetic rats induced single dose aloksan 150 mg/ kg body weight. Rats were divided in to five group (I-V) of six animal in each. The MCE was administrated orally at the dose rate of 100 and 200 mg/kg body weight and compared glibenclamide. Result study that oral administration of both dose of MCE showed significant (p<0,05) decrease in fasting bood glucose compared to diabetic rats. The treatment also resulted increase enzim SOD, GPx concentration and reduced MDA concentration. The result clearly suggest that MCE at higher dose rate (200 mg/kg body weight) more effectively normalize the impaired antioxidant status in aloxan induced diabetes than glibenclamid treated group. The higher dose MCE also more efective in increase improvement in diameter of islet cell of pancreas significant compare with diabetic rats. A significant correlation betwen antioxidan activity (SOD and GPx) MCE with improvement diameter of islet cell in pancreas of aloxan induced diabetic of rats.
