Abstract
Development of herbal drug combination treatment is continously done. The combination is expected to increase the potency of herbal medicine with fewer side effects. This study aims to determine the inhibition effect of anaphylaxis reactions from combination of ethanol extract of black cumin (Nigella sativa L.) seeds and ethanol extract meniran (Phyllanthus niruri L.) herbs, in the ovalbumin-induced rats. This study used an active cutaneous anaphylaxis method with supported histopathology test. Rats were sensitized twice with ovalbumin to induce anaphylaxis reactions, and with evans blue dye to be able measure the diameter of pigmentation on rat backbone. Histopathological observation conduct by toluidine blue, to show the mast cells appear as purple granules on microscopic view. Identification active cutaneous anaphylaxis method used 11 test group, which are the negative control group, the positive control group, the normal control group, 2 a single dose group, and the dosage combination in 6 groups. Data obtained were analysed by ANOVA statistic. The results showed that the combination ethanol ekstract of black cumin seeds and meniran herbs with various doses of the combination, capable of inhibiting the activity of an anaphylaxis reaction in rats. Dose combination of black cumin seed extract: meniran herbs, 15:40.5 mg/kg bw (50%:50%), 22.5:20.25 mg/kg bw (75%:25%), 7.5:60.75 mg/kg bw (25%:75%), 7.5:40.5 mg/kg bw (25%:50%), 7.5:20.25 mg/kg bw (25%:25%), and 22.5:60.75 mg/kg bw (75%:75%) giving % inhibition anaphylaxis reactions, 20.56±4.00; 27.68±5.31; 16.19±4.79; 19.51±5.01; 8.66±2.29 and 17.16±3.04 respectively. Based on histopathological observations, the combination ethanol ekstract of black cumin seeds and meniran herbs inhibition was degranulation of mast cell, and provide a strong correlation, the direction and meaning to the inhibition of active cutaneous anaphylaxis.
