Abstract
Captopril as an angiotensin converting enzyme (ACE) inhibitor is widely used for heart failure and hypertension treatment because of effective and low toxicity. Captopril has a short half-life and suitable for sustained release dosage form. This research aimed to determine the effects and interactions HPMC-Na CMC as matrix to the physical properties of captopril tablet and drug release, and to finding out the optimum proportion of HPMC-Na CMC in captopril sustained release tablet. Simplex lattice design with two variables was applied to optimize the captopril sustained release tablet with matrix HPMC (A) and Na CMC (B). This research was made in three formulas are FI (100% A), FII (75% A : 25% B), FIII (50% A : 50% B), FIV (25% A : 75% B) and FV (100% B) using wet granulation method. The granules and tablets were tested by their physical properties. Flow time, friability, Q60, Q360, and DE360 were used as parameters of optimum formula. The results were computed using software Design Expert version 8.0.6.1. to determine the optimum formula. The optimum formula was compared with predicted value statistically with one sample t test 95% of confidence interval. The results showed that the proportion of HPMC and Na CMC influenced the physical properties granules, tablets and drug release. The interaction between HPMC and Na CMC improved the fluidity and friability, and reduced the drug release (DE360, Q60 and Q360). The optimum formula containing HPMC 70% and Na CMC 30%, with not significant difference between predicted and observed value. Keywords : Captopril, sustained release tablet, HPMC, Na CMC
