Abstract

Cervical cancer is a problem in developing countries. HeLa cell is cervical cancer cells caused by Human papillomavirus (HPV 18) infection so have different properties than normal cervical cells. Treatment of cancer with plants is a very cheap way for the Indonesian population which rich with medicinal plants one of which is a Jombang leaf (Taraxacum officinate Wiggers) plant. This study aims to determine the cytotoxic activity of methanol extract and water fraction of Jombang leaf (Taraxacum officinale Wiggers) in HeLa cells. HeLa cells were suspended at a density of 1 x 104 cell / well as many as 100 µl. The wells containing the cell suspension was added 100 µl test solution i.e the methanol extract and water fraction from the methanol extract of Jombang leaf (Taraxacum officinate Wiggers). Controls used are cell plus media (without addition of test solution) and then the cells were incubated in incubator CO2 5% at temperatur 37°C for 24 hours. Absorbance read using ELISA reader at a wavelength of 595 nm. Data of uptake measurements using MTT data describing total of living cells. IC50 value calculated from the linearity relationship between log concentration to % cell viability. IC50 was a level that can inhibit proliferation by 50% of the cells population. Based on the IC50 obtained from the results of this study concluded that the methanol extracts and the water fraction from Jombang leaf were not potential developed as an agent of cervical anticancer with IC50 value of methanol extract was 51,972 µg/ml and water fraction was 49,9281 ug / ml.
