Abstract
Datura stramonium L. is one of the medicinal plants in Indonesia, included in the Solanaceae family. Kecubung plants contain flavonoid, alkaloid, saponin, polyfenol compounds. Kecubung plants can be used to treat asthma, scabies, edema, eczema. The study was aimed to find out the ability of NAA and BAP growth regulator to induce the callus of kecubung (Datura stramonium L.) leaves, to know the concentration of NAA and BAP growth regulator addition that affected callus formation of kecubung leaves in MS medium, and to know chromatographic profile and the content of compounds in the callus of kecubung (Datura stramonium L.) leaves. The experiment was conducted with plant tissue culture method using Murashige Skoog (MS) medium with the addition of NAA and BAP plant growth regulators, with various concentration of NAA 2 ppm : BAP 0 ppm, NAA 1.5 ppm : BAP 0.5 ppm, NAA 1 ppm : BAP 1 ppm, NAA 0.5 ppm: BAP 1.5 ppm, NAA 0 ppm : BAP 2 ppm. Evaluation of callus was conducted on the percentage of success of explants to form callus, induction time, and the average weight of callus. Analysis of the mother plant and callus was done qualitatively with preliminary test using color reaction and thin layer chromatography. Maceration of mother plant and kecubung leaves callus used sthanol 96%, fractionation was performed using n-hexane, ethylacetate and water. Thin layer chromatography used butanol : acetic acid : water (4:1:5) as mobile phase, and cellulose as stationary phase. The results in mother plant and kecubung leaf (Datura stramonium L.) callus with various concentration of plant growth regulators addition of NAA and BAP gave different chromatographic profiles and different content of compounds in the mother plant and callus of kecubung (Datura stramonium L.) leaves.
