Abstract

Pandanus conoideus Lam is a herbal plant good as anticancer, antidiabetic, antihypertension, and antioxidant. This study was aimed to known the levels of total flavonoid content of each fraction of ethyl acetate subfraction of Pandanus conoideus Lam and to known if there was significant differences in total flavonoid content of each fraction of ethyl acetate subfraction of Pandanus conoideus Lam. Pandanus conoideus Lam contains flavonoids, terpenoids, polyphenols, and alkaloids was extracted using maceration with methanol. The obtained methanol extract was suspended with water, then partitioned with ethyl acetate and n-hexane. fraction of ethyl acetate extract of ethyl acetate fraction and then fractionated by vacuum liquid chromatography. The column was eluted with a combination of mobile phase, and the eluent was collected every 50 ml. Mobile phase combinations used were Petrolium ether: CHCl3: CH3OH. Determination of total flavonoid content performed by spectrophotometry using aluminum chloride reagent in accordance with the procedures by chang et al 2002. The results of column chromatography was 65 subfraction, then the 65 fractions were monitored by TLC. Based on the similarity of Rf values, the merger of the fraction was conducted into 6 major fractions, i.e subfraction A (1-18) , subfraction B (19-30), subfraction C (31-39), fraction D (40-50) 0, fraction E (51 -58), subfraction F (59-65). From the results of this study it was known that the total flavonoid content in whole subfraction A were as follows : 0, subfraction B : 9.108 g / ml, subfraction C : 25.954 ug / ml, subfraction D : 6.919 ug / ml, subfraction E : 3.915 ug / ml, subfraction F : 2,345 ug / ml. It can be concluded that the samples subfraksi C had the highest flavonoid content compared to other subfraction.
